基于UPLC-Q-TOF-MS/MS结合多元统计分析不同产地布渣叶化学成分的差异

摘要:
目的:应用超高效液相色谱-四极杆-飞行时间质谱(UPLC-Q-TOF-MS/MS)技术,结合多元统计分析技术考察不同产地布渣叶的差异性化学成分。方法:色谱条件为Waters CORTECS UPLC C18色谱柱(2.1 mm×150 mm,1.6 μm),以甲醇(A)-0.1%甲酸水溶液(B)流动相进行梯度洗脱,流速0.25 ml/min,进样量1 μl。采用电喷雾电离(ESI)技术,在负离子模式进行采集检测。应用主成分分析(PCA)与正交偏最小二乘法-判别分析(OPLS-DA)等多元统计技术对代谢轮廓及化学成分进行差异性聚类,并筛选布渣叶的差异化学成分。结果:安徽产地与其他产地布渣叶在化学成分上差异有统计学意义。鉴定出13个黄酮类差异成分(3个原花青素类、5个黄酮碳苷类、5个黄酮醇氧苷类)。结论:通过该研究建立的UPLC-Q-TOF-MS/MS多元统计分析方法,明确了不同产地布渣叶的差异化学成分,为布渣叶的质量控制提供了依据,为揭示生态环境对布渣叶代谢物合成积累的影响规律提供了基础资料。

Abstract:
Objective:To investigate the difference in chemical components between Microcos paniculata from different producing areas based on ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS/MS) and multivariate statistical analyses.Methods:UPLC was performed on a Waters CORTECS UPLC C18 column (2.1 mm×150 mm,1.6 μm) with a mobile phase of methanol (A)-0.1% formic acid solution (B) for gradient elution at a flow rate of 0.25 ml/min and a sample size of 1 μL.The electrospray ionization technique was used to perform collection and detection in the negative ion mode.Multivariate statistical analyses,including principal component analysis and orthogonal partial least squares-discriminant analysis,were used to perform differential clustering of metabolic profile and chemical components and screen out the differentially expressed chemical components of Microcos paniculata.Results:There was a significant difference in chemical components between Microcos paniculata from Anhui and that from other producing areas.A total of 13 differentially expressed flavonoids were identified,among which there were 3 procyanidins,5 flavone C-glycosides,and 5 flavonol O-glycosides.Conclusion:By establishing a method of UPLC-Q-TOF-MS/MS combined with multivariate statistical analyses,this study clarifies the difference in chemical components between Microcos paniculata from different producing areas,which provides a basis for the quality control of Microcos paniculata and provides basic data for revealing the influence of ecological environment on the synthesis and accumulation of Microcos paniculata metabolites.
